Ca2+-activated K+ Channels Expressed in Rat Ventricular Fibroblasts

Abstract

|
Cardiac fibroblasts contribute to structural, biochemical, mechanical and electrical properties of the myocardium and form one of the largest cell populations in the heart. However, they are often disregarded by in vivo and in vitro studies into cardiac functions, especially, electrophysiological properties. Ca2+-activated K+ (KCa) channels, which are widely distributed in a variety of cells, can control Ca2+ influx as well as a number of Ca2+-dependent physiological processes. We, therefore, performed a study to identify and characterize KCa channels in rat cardiac ventricular fibroblasts. With immunostaining with discoidin domain receptor 2 (a specific fibroblast marker), we confirmed that the cultured cells from rat heart were fibroblasts and found the expression of various KCa channels (¥á- and ¥â-subunit of BK, IK, and SK3 channels) in reverse transcription polymerase chain reaction. In addition, we identified BK, IK, and SK channels with specific blockers or activators by whole-cell mode patch clamp technique and showed these channels were involved in cell proliferation by MTT assay. From these results, we suggest that the KCa channels may play important role in cell proliferation.
